PCx or Particle Counting of Latex technology is based on microparticles agglutination followed in non equilibrium conditions.Polystyrene microspheres ( Æ of particles is ± 0.8 µm) coated with antigen [or antibody] are strongly mixed with corresponding soluble antibody [or antigen] ; agglutination occurs and after a certain time, the reaction is stopped by massive dilution of the medium.The free particles (non-agglutinated microspheres) are then counted.After the reaction, the number of remaining free particles is inversely proportional to the extent of agglutination.By plotting particle number versus analyte concentration it is possible to generate calibration curves similar to those obtained in other immunoassays.
